    Whole cell extracts (250 µg total protein) prepared from SH-SY5Y cells were immunoprecipitated overnight with 2.0 µg of purified rat IgG2a, κ isotype control (Cat. No. 400502) or purified anti-GATA4 (clone W19078B). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a rabbit control antibody against a separate epitope of GATA4. Description

GATA binding protein 4 (GATA4) is a zinc-finger transcription factor that plays a crucial role in transcriptional activation and cell growth, differentiation, survival, and senescence. It is essential to cardiac development and function. Along with GATA6, it modulates cardiac stress response and regulates cardiomyocyte formation, replication, and function. GATA4 participates in second heart field transcription and differentiation and ventricular morphogenesis through downstream regulation of Mef2c and Hand2. Mutations in GATA4 can result in myocardial abnormalities, myocardial hypoplasia, absence of a proepicardium, abnormal valve formation, and endocardial cushion defects. In the intestine, GATA4, along with GATA6, is also responsible for regulating epithelial cytodifferentiation and structure, and for maintaining balance between enterocytes and secretory cells through the coordination of Notch signaling. GATA4 plays a vital role in human testicular development through its regulation of the expression of steroidogenic factors StAR, INHA, HSD3B2, and CYP19A1 and sex-determining genes SRY, AMH, and SOX9.

Recommended Usage

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC using HeLa cells and SH-SY5Y cells fixed with 4% paraformaldehyde and permeabilized with either methanol or Triton X-100. Both methods resulted in specific, but weak GATA4 staining. We therefore do not recommend this clone for the application.

While this clone detected GATA4 by WB, it also showed a very weak affinity for the protein. Because of this, we do not recommend this clone for the application.

This clone failed IHC testing due to lack of specificity for GATA4. We therefore do not recommend this clone for the application.

This clone is not compatible with our True Nuclear™ Transcription Factor and Cyto-Fast™ buffer sets. We recommend using using True-Phos™ Perm Buffer (Cat. No. 425401) for ICFC.
