Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a .45 µm PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-GSK-3α/β Antibody, clone W17173C, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular Weight marker.

    Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.0 µg of clone W17173C or with Purified Rat IgG2a, κ Isotype Control Antibody (Cat. No. 400502). The resulting IP fractions and whole cell extract input (5%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a .45 µm PVDF membrane, and probed with a separate anti-GSK-3 α/β antibody raised with a different immunizing GSK sequence. Lane M: Molecular Weight marker.

Description

GSK-3α/β is a ubiquitously expressed serine/threonine kinase that can phosphorylate a wide range of substrates. While GSK-3 was initially shown to regulate energy metabolism through phosphorylation of glycogen synthase, it has been shown that the enzyme has a wide variety of substrates in vivo and regulates signaling pathways that control cell growth, development,  apoptosis, and oxidative stress responses. Each isoform if encoded by separate alleles; the isoforms show 66% homology.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone recognizes both a and ß isoforms of GSK-3 by both WB and IP.

This clone failed to stain PFA-fixed HeLa cells that were permeabilized with methanol or Triton-X. We therefore do not recommend using this clone for ICC.
