    Human paraffin-embedded placenta tissue slices were prepared with a standard protocol of deparaffination and rehydration. Antigen retrieval was done with citrate buffer 1X (10 mM, pH6.0) at 95°C for 40 minutes. The tissue was washed with PBS/ 0.05% Tween-20 twice for five minutes and blocked with 5% FBS and 0.2% gelatin for 30 minutes. Then, the tissue was stained with 5 µg/mL purified anti-human CD235ab recombinant (clone QA20B11) at 4°C overnight. On the next day, the tissue was washed twice with PBS and stained with Alexa Fluor® goat anti-mouse IgG (clone Poly4053) (red) for two hours at room temperature. The nuclei were counterstained with DAPI (blue). The image was captured with a 10X objective.

Description

The clone QA20B11 antibody reacts with a common epitope of glycophorin A (CD235a) and glycophorin B (CD235b). Glycophorin A is a major sialoglycoprotein expressed on red blood cell membrane, and erythroid precursors. Glycophorin A shares strong homology with glycophorin B. The clone QA20B11 antibody recognizes human RBCs and erythroid precursors and is useful in erythroid cell development studies. Mature, non-nucleated red blood cells are characteristically glycophorin A positive, but CD45 and CD71 negative.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.001 µg per million cells in 100 µL volume. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 2.5 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
