Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL of purified anti-KDM1A (clone W20070A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.5 µg of purified rat IgG2a, κ isotype control (Cat. No. 400502) or purified anti-KDM1A (clone W20070A). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with a rabbit control antibody against a separate epitope of KDM1A. IHC staining of purified anti-KDM1A (clone W20070A) on formalin-fixed paraffin-embedded human testis tissue. Following antigen retrieval using 1X Tris-EDTA buffer (10mM Tris, 1mM EDTA, pH 9.0), the tissue was incubated without (panel A) or with 1 µg/mL of anti-KDM1A (panel B), overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-rat IgG (red) (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and images were captured with a 60X objective. Scale bar: 50 μm -
HeLa cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with Triton X-100 for 10 minutes, and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with 1.5 µg/mL of either purified rat IgG2a, κ isotype control (Cat. No. 400502) (panel A) or purified anti-KDM1A (clone W20070A) (panel C) overnight at 4°C followed by incubation with of Alexa Fluor® 594 goat anti-rat IgG (Cat. KDM1A (or LSD1) belongs to the flavin adenine dinucleotide (FAD)-dependent demethylase family and has enzymatic activity on de-methylation lysine-4 and lysine-9 of Histone H3. During early stage embryonic development, KDM1A regulates differentiation and development through corepressor of RE1-silencing transcription factor (CoREST) complex and histone deacetylases (HDACs). KDM1A participates in nuclear hormone signaling by interacting with androgen receptor (AR) and estrogen receptor (ER) of the receptor-expressing tissues in the regulation of cancer progression and metastasis. In addition to histone H3, KDM1A demethylates lysine residues of p53, E2F1, or DNMT1 which modulates apoptosis and cell proliferation. Dysfunction of KDM1A is found in acute myeloid leukemia (AML), T cell acute lymphoid leukemia (T‑ALL), and several malignancies, implying it is a potential epigenetic biomarker and drug target.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Clone W20070A does not cross-react with mouse in western blot.
For immunocytochemistry (ICC), the suggested fixation/permeabilization method is either 4% paraformaldehyde/0.5% Triton X-100 or paraformaldehyde followed by ice-cold methanol.
