    Mouse NIH/3T3 fibroblast cells were fixed with 4% PFA Fixation Buffer, washed with PBS and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with purified anti-mouse DLK1 (clone W21075D, panel A) or 2.5 µg/mL purified rat IgG2b, κ isotype control (panel B) followed by 2.5 µg/mL of Alexa Fluor 555® anti-mouse IgG (Cat. No. 405324) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Images were captured with a 40x objective. Scale bar: 20 µm

Description

The Delta-Notch pathway controls developmental processes including cell fate determination, terminal differentiation, and proliferation. In mammals, four Notch receptors (NOTCH1–4) and five activating canonical ligands (JAGGED1, JAGGED2, DLL1, DLL3 and DLL4) have been described. Delta-like 1 homolog (DLK1) is a noncanonical Notch ligand, and acts as an inhibitor of Notch signaling. The inhibitory effect of DLK1 and DLK2 on Notch signaling may be mediated by competition with canonical ligands for the receptor binding site. Widely expressed during embryonic development, expression becomes restricted in the adult to neuroendocrine tissues like the pituitary gland, adrenal glands, pancreas, monoaminergic neurons in the central nervous system, testes, prostate, and ovaries.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 2.5 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
