Total cell lysates (15 µg total protein) from HeLa and Molt-4 (reduced expression controls), Daudi, and A20 cells were resolved by 4-20% Tris-glycine gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti-OCT2 Antibody, clone 2B10B19, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-Actin Antibody (Cat. No. 643807) was used as a loading control at a 1:5000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

Description

OCT2 is a member of the POU family of transcription factors that was found to be expressed predominantly in B lymphocytes, activated T lymphocytes, and neuronal cells. OCT2 binds to the conserved octamer DNA element, ATGCAAAT, which was discovered in the promoters and enhancers of immunoglobulin (Ig) genes. In addition to regulating Ig gene expression, OCT2 controls several B cell specific genes required for proliferation and differentiation, such as CD20, CD36, and CD79a. OCT2 was found to have different isoforms produced by alternative splicing. The OCT2 isoforms exert distinct functional activities and can act as transcriptional activators or repressors.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone may produce a non-specific band of ~75 kD by western blot.
