SDHB encodes one of the four proteins that form the SDH (succinate dehydrogenase) enzyme complex in the mitochondrial-respiratory chain, and plays an important role in the mitochondria. The SDH complex functions to couple the conversion of succinate to fumarate in the mitochondrial matrix as part of the Krebs cycle with the reduction of ubiquitin to ubiquinol as part of the ETC (electron transport chain). Within the complex itself SDHB provides an attachment site for released electrons to be transferred to the oxidative phosphorylation pathway. Sporadic and familial mutations in this gene result in paragangliomas, pheochromocytoma, and gastrointestinal stromal tumors.

- Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Clone W19244A is suitable for ICC using methanol only as the fixation/permeabilization method. Alternate methods such as fixation with PFA followed by permeabilization with either methanol or Triton-X-100 resulted in staining in the wrong localization.
