Total protein from human brain (5 µg, lane 1), mouse brain (10 µg, lane 2), rat brain (10 µg, lane 3) and 15µg of total protein from NTERA2 (lane 4), HeLa (lane 5), EL-4 (lane 6) were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:500 diluted (1 µg/mL), 1:1000 diluted (0.5 µg/mL) and 1:2000 diluted (0.25 µg/mL) purified anti-SIRT2 antibody (upper). Proteins were visualized using an HRP anti rat-IgG secondary antibody (Cat. No. 405405) or HRP Donkey anti-rabbit IgG Antibody (Cat. No. 406401) for GAPDH and chemiluminescence detection. GAPDH (poly6314, Cat. No. 631401) antibody was used as a loading control (lower). Lane M: Molecular weight ladder.

Description

Mammalian sirtuin family proteins are homologs of the Saccharomyces cerevisiae silencing protein Sir2. Sirtuins are NAD-dependent deacylases and ADP-ribosyl transferases involved in the regulation of various cellular processes, such as metabolism, autophagy, apoptosis, gene silencing, and lifespan. Unlike the other sirtuin members, Sirtuin 2 (SIRT2) is predominantly distributed in the cytoplasm and functions as a microtubule deacetylase. SIRT2 also deacetylates lysines on histone and several key transcriptional factors, including P53 and NF-κB. SIRT2 shuttles continuously between cytoplasmic and nuclear during interphase, associates with chromatin in early prophase, and localizes to centrosomes and the mitotic spindle during metaphase. SIRT2 has been implicated in the pathology of cancer and neurodegenerative diseases. SIRT2 deacetylates histone proteins and prevents chromosomal instability during mitosis. Inhibition of SIRT2 has been shown to negatively regulate polyglutamine inclusion and rescue neuronal cells from α-synuclein-induced toxicity.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.25 - 1.0 µg per ml (1:500 - 1:2000). It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

As per the Human Protein Atlas RNA level dataset, NTERA2 cell is expected to have a low level of SIRT2 expression.

Human SIRT2 has 5 isoforms with Mw ranging from 30kDa to 43kDa.

This clone cannot be used for IF application (in house tested).
