The transcription regulator SMARCA4/BRG1 is part of the SWI/SNF protein family and regulates chromatin structure in an ATP-dependent manner. It has helicase and ATPase activities which are required for chromatin topology remodeling. SMARCA4/BRG1 is important in transcriptional regulation of several genes involved in stress responses, neurogenesis, cell cycle, and differentiation. In this context, transcription of c-FOS in resting neurons is inhibited by recruitment of phospho RB1-HDAC complex by SMARCA4/BRG1. Alternatively, the CREST-BRG1 complex induces NR2B expression by binding to its promotor. In addition, SMARCA4/BRG1 regulates the expression of the tumorigenic protein CD44. Lastly, mutations in the SMARCA4 gene have been associated with rhabdoid tumor predisposition syndrome type 2.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1 - 5 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For intracellular flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
