Whole cell extracts (15 µg total protein) from serum starved A431 cells untreated (-) or treated (+) with IFN-α (10 ng/mL, 30 min) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1 µg/mL of purified anti-STAT2 phospho (Tyr690) (clone W21151B) for 2 hours at room temperature. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 649203) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Description

STAT2 is a member of the STAT family of transcription factors that is phosphorylated by receptor associated kinases in response to cytokines and growth factors. After activation, STAT2 forms heterodimers with STAT1. Once it is activated, STAT1 and STAT2 dimerize and are translocated to the nucleus to act as a transcription factor. STAT2 is an important mediator of type I interferon signaling; thus, after type-I IFN binds to the receptor, JAK kinases phosphorylate STAT2 and STAT1. This leads to their dimerization and, together with IRF9/ISGF3G, form the ISGF3 transcription factor. Type-I IFN /STAT2 signaling is important in innate immunity antiviral responses. Additionally, STAT2 is a regulator of mitochondrial fission by modulating phosphorylation of DNM1L. Diseases associated with STAT2 are the Immunodeficiency 44 and Pseudo-Torch Syndrome 3.
