Description

VPS35 is the largest subunit of the retromer complex, which plays an important role in retrograde protein transport from the endosome to the trans-Golgi network. VPS35 binds to VPS26 (VPS26A or VPS26B) and VPS29 to form the vacuolar protein sorting subcomplex (VPS), which in turn interacts with different nexins to form different retromer variants that transport different targets. D620N mutation in VPS35 has been implicated as a causative genetic factor for developing late-onset, autosomal dominant familial Parkinson’s disease, and could serve as potential therapeutic target for PD.
