TGF-β1 and its receptors play an important role in normal development and homeostasis. TGF-β ligands (TGF-β1, TGF-β2, TGF-β3) bind to three receptor isoforms TGFBR1, TGFBR2, and TGFBR3. TGFBR1 and 2 are serine/threonine and tyrosine kinases; TGFBR3 does not possess kinase activity. The TGFBR1 and TGFBR2 have similar structure characterized by a cysteine rich extracellular domain, a transmembrane helix, and the intracellular serine-threonine kinase domain. The TGFBR2 binds with high affinity to the ligands, and this binding promotes the recruitment and transphosphorylation of TGFBR1. TGFBR2 is also present in soluble forms named TβRII-SE splice variant. This variant binds to the three TGFβs with high affinity. TGFBRII-SE has been used in a model of liver fibrosis, and overexpression of TGFBRII-SE-Fc decreases liver damage and reduces liver fibrosis. TGF-β1 and TGFBR2 play a key role in suppressive and inflammatory immune responses. TGF-β controls thymic T-cell selection and homeostasis of naïve T-cell. TGF-β1 promotes peripheral Treg, Th9, Th17, and Tfh cell generation. In addition, TGF-β1 suppresses CTL, Th1, and Th2 cells.

- Recombinant human TGFBR2-Fc Chimera, amino acid sequence containing Thr23-Asp159 (Accession No. P37173), was expressed in 293E cells. The carboxy-terminus contains SR-IEGRMD-hIgG1 (Pro100-Lys330)-Fc tag.
- Molecular Mass
- The 376 amino acid recombinant protein has a predicted molecular mass of approximately 42.4 kD. The DTT-reduced and non-reduced proteins migrate at approximately 60 kD and 100 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Thr.
- Purity
- > 95%, as determined by Coomassie stained SDS-PAGE.

- 0.22 µm filtered protein solution is in 10 mM Na2HPO4, 0.3 M NaCl, pH 7.2
- Endotoxin Level
- Less than 0.1 EU per µg cytokine as determined by the LAL method.
- Concentration
- 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
- Activity
- Immobilized recombinant human TGF-β1 (Cat. No. 781802) binds to human TGFBR2-Fc in a dose dependent manner. The ED50 for this effect is 2 – 10 ng/mL.

- What is the specific activity or ED50 of my recombinant protein?

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
