Anti-human GDNF antibody (purple squares, clone A16099G) inhibits the binding in a dose-dependent manner while the isotype control (grey triangles, Cat. No. 401215) does not have the effect. This antibody blocks the binding of 0.1 µg/mL recombinant human GFRα1 protein to 0.5 µg/mL of immobilized recombinant human GDNF with an ND50 = 0.05 - 0.4 µg/mL.

Description

Glial cell-derived neurotrophic factor (GDNF) is a member of the TGF-β superfamily and was first identified as a growth factor supporting the survival of embryonic midbrain neurons. GDNF has been shown to promote the development, differentiation, and survival of dopaminergic neurons by signaling through GDNF family receptor α-1 (GFRα1) and the coreceptor RET receptor tyrosine kinase. Dysregulated expression of GDNF has been reported to be associated with several mental diseases, which include depression and schizophrenia. GDNF is considered as a promising therapeutic agent for the treatment of Parkinson's disease because it can protect dopaminergic neurons from neurotoxins and promote neuron regeneration after injury. In addition to the functions in the nervous system, GDNF is also required for the differentiation of spermatogonia and kidney development. Upregulated expression of GDNF was observed in various cancer cell lines and correlates with malignant cancer progression, suggesting a role of GDNF in carcinogenesis.

The Ultra-LEAF™ (Low Endotoxin, Azide-Free) antibody was purified by affinity chromatography.
Concentration
The antibody is bottled at the concentration indicated on the vial, typically between 2 mg/mL and 3 mg/mL. Older lots may have also been bottled at 1 mg/mL. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C. This Ultra-LEAF™ solution contains no preservative; handle under aseptic conditions.

Recommended Usage

Each lot of this antibody is quality control tested by blocking the binding between recombinant human GFRα1 and recombinant human GDNF (Cat. No. 760402). The ND50 range is 0.05 - 0.4 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.
