Total cell lysate (15 µg protein) from HeLa (negative control), HaCaT, A431 and NIH3T3 cells were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1:1000 (0.5 µg/mL) diluted purified anti-Cytokeratin 6 (clone W17006B, upper). Proteins were visualized by chemiluminescence detection using 1:3000 diluted HRP rat-IgG secondary antibody (Cat. No. 405405). 1:2000 dilution of Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1, Cat. No. 643807) was used as a loading control (lower). Lane M: MW ladder. -
HaCaT cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with 1:500 diluted (1 µg/ml, Figure A) rat IgG2b isotype control (Cat. No. 400601)or 1:500 (1 µg/ml, Figure B), 1:2000 (0.25 µg/ml, Figure C) and 1:5000 (0.1 µg/ml, Figure D) diluted anti-Cytokeratin 6 overnight at 4° followed by 1:250 diluted (2 µg/ml) Alexa Fluor® 488 (green) conjugated goat anti-rat IgG (Cat. No. 405418) antibody for one hour at room temperature. Nuclei were counterstained with DAPI (blue). The image was captured with a 60X objective. -
IHC staining using purified anti-cytokeratin 6 antibody (clone W17006B) on formalin-fixed paraffin-embedded human tonsil tissue. Following antigen retrieval using Tris-EDTA buffer (10mM Tris, 1mM EDTA, pH 9.0), the tissue was incubated with 10 µg/mL of antibody overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 goat anti-rat IgG antibody (magenta) (Cat. No. 405416) for one hour at room temperature. Nuclei were counterstained with DAPI (blue) (Cat. No. 422801), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 20X objective. Scalebar = 50 µM.

Keratins are main structural proteins of epithelial cells and form a cytoplasmic intermediate filament network, which is required for cell integrity and protecting cells from stress or injury. Keratin 6 (KRT6), also known as cytokeratin-6 or CK-6, is a type II intermediate filament protein constitutively expressed in all major types of epithelial appendages, such as hair, nail, and glands. Expression of KRT6 is induced by cell injury, infection, and disease. KRT6 has been identified as a marker of mammary gland progenitor cells. Defects in KRT6 genes have been associated with the inherited disorder pachyonychia congenital (PC), leading to structural abnormalities of nail, palms, and soles.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/ml (1:500-1:5000 dilution). For immunocytochemistry, a concentration range of 0.1 - 1.0 µg/ml (1:500-1:5000 dilution) is recommended. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
