Total lysates (15 µg protein) from HepG2, Daudi and UMR106 (rat) were resolved by electrophoresis (4-20% Tris-glycine gel), transferred to nitrocellulose, and probed with 1 µg/ml purified anti-hnRNPK antibody (clone F45P9C7). Proteins were visualized using chemiluminescence detection by incubation with HRP goat anti-mouse IgG secondary antibody (Cat. No. 405306, 1:3000 dilution). Direct-Blot™ HRP anti-β-actin was used as a loading control (Cat. No. 643807, 1:8000 dilution). -
Total cell lysate from HeLa cells (lane 1, 15 µg), 293E cells (lane 2, 15 µg), U87MG (lane 3, 15 µg), mouse brain (lane 4, 15 µg) and rat brain (lane 5, 15 µg) were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with purified anti-hnRNPK antibody (clone F45P9C7). Proteins were visualized using an HRP Goat anti-mouse IgG Antibody and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1) was used as a loading control.

hnRNP K (Heterogeneous nuclear ribonucleoprotein K) is a pre-mRNA binding protein and a component of hnRNP complexes providing substrate for pre-mRNA processing before becoming translatable mRNAs in cytoplasm. This protein containing KH domains and is ubiquitously expressed in the cytoplasm and the nucleus with a predicted molecular weight approximately 51 kD. hnRNP K expression and function are increased by epidermal growth factor (EGF). hnRNP K acts as transcriptional regulator and functions in nucleocytoplasmic shuttling, and is modified by phosphorylation (PKC δ, c-Src) and acetylation. Clone F45P9C7 has been shown to be useful for western blotting and immunohistochemistry of human, mouse, rat, rabbit and chicken hnRNP K.
