The human RXR (retinoid X receptor) includes three distinct genes (RXRα, β, and γ). RXR is composed of two major domains of nuclear receptor family proteins, the DNA-binding domain (DBD) and the ligand binding domain (LBD) that bind with high affinity to 9-cis –RA (Retinoic Acid). The RAR/RXR heterodimers bind to RA response elements (RARE) composed of two 5'-AGGTCA-3' sites to modulate RA-mediated gene expression.
The RXRα, also known as NR2B1, plays a crucial role in various biological effects, such as UVR-induced DNA damage, cell growth, development, metabolism, differentiation, and apoptosis. RXRα is highly expressed in liver and also expressed in kidney and brain. It regulates many intracellular receptor signaling pathways via interacting with ligands and co-regulators. The RXRα/PPARα heterodimer is essential for energy balance regulation such as PPARα transcriptional activity on fatty acid oxidation genes including ACOX1 and the P450 system genes. Proteolytic cleavage of RXRα (tRXRα) protein results in a truncated product found in many tumor cells. The tRXRα (RXRα-Δ80) interacts with the p85α subunit of the PI3K/AKT pathway and leads to the increase of tumor cell proliferation.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

The clone does not cross-react with mouse and rat RXRα.
For ICC, we recommend 4% PFA fixation followed by permeabilization with 0.5% Triton X-100 or ice-cold methanol. We do not recommend ice-cold methanol for fixation and permeabilization.
