    IHC staining of Purified anti-TUBB2B (clone 7B9.rec) on formalin-fixed paraffin-embedded human colon tissue. Following antigen retrieval using Tris-EDTA pH 9.0 (10 mM Tris, 1 mM EDTA), the tissue was incubated without (panel A) and with (panel B) 1 µg/mL of Purified anti-TUBB2B (clone 7B9.Rec) overnight at 4°C. This was followed by incubation with 2.5 µg/mL of Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). Description

Tubulin is a major component of microtubules. It exists as a heterodimer consisting of an alpha and beta subunit. Class II beta tubulin is the major form of tubulin beta in neurons and is found in many other tissues including lung tissue and Schwann cells. TUBB2B is also indicated in various cancers and has been shown to have increased expression in hepatocellular carcinomas.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For the use of this antibody (clone 7B9.rec) in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval with either Tris-EDTA (10 mM Tris, 1 mM EDTA, pH 9.0) or Citrate Buffer, 10X (Cat. No. 420902).

For the use of this antibody (clone 7B9.rec) in immunocytochemistry (ICC), it is recommended to perform fixation/permeabilization with either 100% ice-cold methanol or fixation with Fixation Buffer (Cat. No. 420801) followed by permeabilization with 100% ice-cold methanol.
