Poly5200_LEAF_SCF_Block_022625
Recombinant human CD117-Fc chimera (black circles) binds to immobilized human SCF (Cat. No. 573904). LEAF™ purified anti-human SCF (Poly5200) (purple squares) inhibits the binding in a dose-dependent manner, whereas the LEAF™ purified goat IgG isotype control (gray triangles) does not have the effect. This antibody blocks the binding of 0.025 µg/mL human CD117-Fc chimera to 1 µg/ml immobilized human SCF. The ND50 range: 0.04 - 0.3 µg/mL

Description

Stem Cell Factor (SCF) was initially synthesized as membrane-bound protein of 248 or 220 amino acids, depending on alternative splicing of exon 6. The 248 amino acid form contains a proteolytic cleavage site encoded by exon 6 which is cleaved from the cell to release an active soluble protein of 165 amino acid residues. Soluble SCF is glycosylated at both N-linked and O-linked sites. MMP-9 plays a physiological role in SCF release from the membrane, and this action plays a significant role in differentiating and mobilizing stem and progenitor cells from the bone marrow. SCF increases the proliferation of myeloid and lymphoid hematopoietic progenitors in bone marrow cultures. SCF/c-kit interaction in mast cells results in mast cell degranulation with release of mediators, such as histamine and inflammatory cytokines and chemokines. Activation of c-kit in dendritic cells regulates T helper cell differentiation and allergic asthma. In addition, SCF plays an important role in re-vascularization of ischemic limbs. Ischemia induces plasma elevation of SCF and thrombopoietin (TPO) and, in lower levels, GM-CSF and erythropoietin (EPO). SCF and TPO can induce the release of CXCL12 from platelets into plasma. This results in an extensive mobilization of CXCR4+VEGFR1+ cells (hemangiocytes), accelerating re-vascularization of the ischemic limbs. SCF binds to its receptor kit that belongs to the type III tyrosine kinase family, whose members include receptors for M-CSF and PDGF.

Application

Block - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by the ability to block the binding of an experimentally-determined spike concentration of human CD117-Fc to immobilized human SCF (Cat. No. 573904)   The ND50 range for this effect is 0.04 - 0.3 µg/mL.  It is recommended that this reagent be titrated to confirm performance.

Related FAQs

Do you guarantee that your antibodies are totally pathogen free?

BioLegend does not test for pathogens in-house aside from the GoInVivo™ product line. However, upon request, this can be tested on a custom basis with an outside, independent laboratory.

Does BioLegend test each Ultra-LEAF™ antibody by functional assay?

No, BioLegend does not test Ultra-LEAF™ antibodies by functional assays unless otherwise indicated. Due to the possible complexities and variations of uses of biofunctional antibodies in different assays and because of the large product portfolio, BioLegend does not currently perform functional assays as a routine QC for the antibodies. However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.

Does BioLegend test each Ultra-LEAF™ antibody for potential pathogens?

No, BioLegend does not test for pathogens in-house unless otherwise indicated.  However, we can recommend an outside vendor to perform this testing as needed.

Have you tested this Ultra-LEAF™ antibody for in vivo or in vitro applications?

We don't test our antibodies for in vivo or in vitro applications unless otherwise indicated. Depending on the product, the TDS may describe literature supporting usage of a particular product for bioassay. It may be best to further consult the literature to find clone specific information.
