Whole cell extracts (15 µg total protein/lane) from HEL cells spiked with 20 ng/lane of Avi-tagged VEGF-165 biotinylated or non-biotinylated recombinant protein, were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.5 µg/mL of purified anti-Avi-tag (clone W19258A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP anti-rat IgG (Cat. No. 405405) at a 1:3000 dilution. Western-Ready™ ECL Substrate Premium Kit (Cat. Whole cell extracts (300 µg) total protein prepared from untransfected or VEGF-165-Avi-tag transfected CHO cell extracts were incubated overnight with 2 µg of purified rat IgG2b, κ isotype control or purified anti-Avi-tag (clone W19258A). The resulting IP fractions and whole cell extract input (6%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to PDGF membrane and probed with a different Avi-tag. -
CHO cells untransfected or transfected with an expression vector for VEGF-165-Avi-Tag were fixed with 4% paraformaldehyde, permeabilized with 0.5% TritonX-100, and blocked with 5% FBS for 1 hour. Cells were then intracellularly stained with purified anti-Avi-Tag (clone W19258A) overnight at 4°C, followed by incubation with Alexa Fluor® 647 anti-Rat IgG2b (Cat. No. 408208) at 2.0 µg/mL. Nuclei and actin filaments were counterstained with DAPI (Cat. No. 422801) and Flash Phalloidin™ Green 488 (cat. No. 424201). The images were captured in a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm. -
CHO cells transfected with an expression vector for VEGF-165-Avi-tag (filled histogram), and untransfected cells (solid line, open histogram) were fixed and permeabilized using the Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) and intracellularly stained with purified anti-Avi-tag (solid line histograms) or purified rat IgG2b, κ isotype control (dashed line, open histogram) followed by PE goat anti-rat IgG.

Avi-tag is a 15 amino acid sequence, and the minimal acceptor peptide of a larger protein domain that serves as a substrate for the biotin ligase BirA from E. coli. Although one of the major advantages of Avi-tag fusion proteins is a controlled biotinylation in this domain, Avi-tag is also a useful tool in cell and molecular biology.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.
