IHC staining of purified anti-Copper ATPase 1 antibody (clone L60/4) on formalin-fixed paraffin-embedded human skin. Following antigen retrieval using 50 mM Tris-buffered saline with Tween 20 (Cat. No. 925501), the tissue was incubated with 10.0 µg/mL of either purified mouse IgG2b, κ isotype antibody (Cat. No. 402202) (panel A) or primary antibody (panel B) overnight at 4°C, followed by incubation with Alexa Fluor® 647 goat anti-mouse IgG (red) (Cat. No. 405322) at 2.5 µg/mL. Nuclei were counterstained with DAPI (Cat. No. 422801), and the slides were mounted with ProLong™ Gold Antifade Mountant. Images were captured with a 40X objective. Scale: 50 µm.

Description

Copper ATPase 1, also known as ATP7A, is a copper-transporting ATPase which uses ATP hydrolysis to transport copper ions across membranes to regulate intracellular copper levels. As copper is an important trace nutrient required for cellular health, ATP7A is widely expressed in most tissues. For example, ATP7A expression in enterocytes in the small intestine controls dietary uptake of copper from digested food. Disruptions to the ATP7A gene result in Menkes disease, whose symptoms are caused by copper deficiency.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
