    HeLa cells were fixed with 4% paraformaldehyde (PFA) for 15 minutes, permeabilized with 0.5% Triton X-100 for 3 minutes, and blocked with 5% FBS for 60 minutes. Then the cells were intracellularly stained with (A) Purified Rat IgG2a, κ Isotype Ctrl Antibody (Negative, Cat. No. 400502) or (B-D) Purified anti-EIF4A1 antibody (clone W17130C) overnight at 4°C. Nuclei were counterstained with DAPI (Blue, Cat. No. 422801). The image was captured with a 60X objective using KEYENCE BZ-X700 fluorescence microscope. Exposure time (Seconds) for (A-D) is 1/30. Concentrations for (A-B) is 8 µg/mL, (C) is 2 µg/mL, (D) is 0.5 µg/mL and (E) is 0.1 µg/mL.

Following antigen retrieval using 70% formic acid for 20 minutes at room temperature, the tissue was incubated with (A, B) or without (C, D) 5 µg/mL of the primary antibody overnight at 4°C. BioLegend´s Ultra-Streptavidin (USA) HRP kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. EIF4A1 is an ATP-dependent RNA helicase. It can form the eIF4F complex, which is involved in translation initiation by binding with the 5' m7 GTP cap of the mRNA. It could unwind mRNA secondary structure at the 5’ UTR region and lead to the small ribosomal subunit binding and followed scanning of the initiator codon.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.025 - 0.5 µg/mL (1:1000 - 1:20000 dilution). For immunocytochemistry, a concentration range of 0.5 - 2.0 μg/mL (1:250 - 1:1000 dilution) is recommended. For immunoprecipitation, the suggested use of this reagent is 5.0 - 15 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 5.0 - 10 µg/mL (1:50 - 1:100 dilution) is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
