Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain membrane lysate; Lane 3: 20 µg of mouse brain membrane lysate; Lane 4: 20 µg of rat brain membrane lysate. The blot was incubated with 1 µg/mL of the primary antibody overnight at 4°C, followed by incubation with HRP goat anti-mouse IgG antibody (Cat. No. 405306).     ICC staining of purified anti-GlyT2 (SLC6A5) antibody (clone A18174E) on primary mouse neurons. The cells were fixed with 4% PFA, permeabilized with a buffer containing 0.1% Triton X-100 and 0.25% BSA, and blocked with 2% normal goat serum and 0.02% BSA. The cells were then incubated with 10 µg/mL of the primary antibody overnight at 4°C, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG for one hour at room temperature. The cells were co-stained with Alexa Fluor® 488 anti-Tubulin β 3 (TUBB3) antibody (clone TUJ1, Cat. No. 801203). The slide was mounted with fluoromount G with DAPI. The image was captured with a 60X objective. Scale bar: 20 µm

Description

Glycine transporter 2 (GlyT2) protein is encoded by the SLC6A5 gene. It is one of the major glycine transporters that is expressed predominantly in the brain stem and spinal cord, and is frequently used as cellular marker for glycinergic neurons in the central nervous system. Mutations in the SLC6A5 gene have been clinically linked to hyperekplexia 3, which has been thought to be caused by disruption of inhibitory neuronal transmission.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 1 - 5 µg/mL. For immunocytochemistry, a concentration range of 2 - 10 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
