Major histocompatibility complex (MHC) class I molecules present peptide antigens recognized by CD8+ T cells to enable the immune recognition of virally or malignantly transformed cells exposing non-self peptides. Selection and loading of peptides onto MHC I is orchestrated by the peptide-loading complex (PLC). Tapasin, a central component of the PLC, stabilizes MHC I and catalyzes the exchange of low-affinity against high-affinity, immunodominant peptides. In this quality-control step, tapasin cooperates with other more general, ER-resident chaperones, including ERp57 and TAP. Tapasin has been found to be downregulated in glioblastoma, renal cell carcinoma, prostate cancer, urothelial carcinoma, cervical carcinoma, head and neck carcinoma and esophageal squamous cell carcinoma. Studies also reported reduced tapasin to be linked to metastasis and poor outcome.

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

This clone does not cross-reacts with human (in-house tested). This antibody recognizes a non-specific band over 100 kD in some tested cell lysates.
