Western blot of purified anti-TFEB antibody (clone A17106C). Lane 1: Molecular weight marker; Lane 2: 20 µg of human brain lysate; Lane 3: 20 µg of mouse brain lysate; Lane 4: 20 µg of rat brain lysate. The blot was incubated with 5 µg/ml of the primary antibody overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse IgG (Cat. No. 405306). Enhanced chemiluminescence was used as the detection system.

IHC staining of purified anti-TFEB antibody (clone A17106C) on formalin-fixed paraffin-embedded human renal carcinoma tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with 5 µg/ml of the primary antibody overnight at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit (Multi-Species, DAB, Cat. No. 929901) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale Bar: 50 µm

Description

The autophagy-lysosomal pathway is involved in the degradation of long-lived proteins. The transcription factor EB (TFEB) plays a critical role in regulating basic cellular processes, such as lysosomal biogenesis and autophagy. The subcellular localization and activity of TFEB are regulated by mechanistic target of rapamycin (mTOR)-mediated phosphorylation. TFEB is also involved in physiological processes, and it induces the intracellular clearance of pathogenic factors in a variety of diseases, such as Parkinson's and Alzheimer's, suggesting that novel therapeutic strategies could be based on the modulation of TFEB activity.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 5.0 µg per ml. For immunohistochemistry on formalin-fixed paraffin-embedded tissue, a concentration range of 5 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Antigen Details

Structure
TFEB is a 476 amino acid protein with a molecular weight mass of approximately 53kD.
Distribution

Tissue distribution: TFEB is expressed in numerous cell types of the central nervous system (CNS).

Cellular distribution: Cytosolic and nucleus.

Function
TFEB is a major regulator of autophagy and lysosomal biogenesis.
Interaction
TFEB interacts with the mTOR complex and its activity is thought to be regulated through the phosphorylation by mTOR.
