Stability Testing for recombinant human Decorin. Recombinant human Decorin was aliquoted in PBS, pH 7.2 at 0.2 mg/mL. One aliquot was kept at 4°C (Control), and another was frozen and thawed four times (4x Freeze/Thaw). The samples were evaluated in a binding assay with immobilized human TGF-β1 at 5 µg/mL and different concentration of recombinant human Decorin that binds in a dose-dependent manner. The ED50 for this effect is 0.07 – 0.49 µg/mL.

A member of the small leucine-rich proteoglycan (SLRP) family, Decorin, also known as PGS2, is a small extracellular chondroitin/dermatan sulfate proteoglycan. Decorin consists of single glycosaminoglycan chain, carboxy-terminal domains, glycosaminoglycan chains, core protein domains, and amino-terminal domains. Decorin is primarily expressed in fibrous connective tissue and involves collagen fiber formation in the tendons, cornea, dermis, and cartilage. Decorin is synthesized and secreted by fibroblasts. Decorin is a component of connective tissues and plays key roles in the regulation of angiogenesis and matrix assembly. Decorin mediates endothelial cell proliferation and migration by interacting with type I collagen fibrils, α2β1 integrin, insulin-like growth factor-I receptor (IGF-IR), vascular endothelial growth factor receptor 2 (VEGFR2), MET, and transforming growth factor β1 (TGF-β1). In its soluble form, Decorin can inhibit cancer growth. Decorin triggers an acute inflammatory response by binding to TLR2 and TLR4 on the surface of macrophages. Decorin enhances the production of proinflammatory factors and activates inflammatory pathways of p38, ERK1/2, and NF-kB pathways.

- Molecular Mass
- The 358 amino acid recombinant protein has a predicted molecular mass of approximately 39.8 kD. The DTT-reduced and non-reduced proteins migrate at approximately 45 - 55 kD by SDS-PAGE. The N-terminal amino acid is Gly.

- > 95%, as determined by Coomassie stained SDS-PAGE.

- 0.22 µm filtered protein solution is in PBS, pH 7.2

- Storage & Handling
- Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.

The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
- Have your recombinants been tested for stability?

Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
