Total cell lysates (15 µg total protein) from HEK293T cells and untreated (-) or treated (+) with UV radiation were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of Purified anti- TIF1β (KAP-1, TRIM28) Phospho (Ser473) Antibody, clone 11G10SC, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG Antibody (Cat. No. 405306) at a 1:3000 dilution. Purified anti-TIF1β (KAP-1, TRIM28) Antibody, clone 20A1 (Cat. 619302), was used as a pan TIF1β loading control at 1:50000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Description

Transcriptional intermediary factors (TIFs) are a group of transcriptional coactivators and corepressors that regulate gene expression by modulating chromatin structure and assembly of transcription initiation complexes. TIF1β is a member of the TIF1 subfamily of chromatin-associated TIFs that play a key role in many developmental and physiological processes. Studies using knockout mice reveal the important function of TIF1β in regulating genomic imprinting, T cell activation, and T cell tolerance.

Structure
An N-terminal RING-B box coiled coil (RBCC) domain most likely attributes to intermolecular structure, while two characteristic chromatin-targeting domains, a plextrin homology (PH) finger motif and bromodomain, are found in the C-terminus.

Function
TIF1β acts as a transcriptional repressor by modulating histone deacetlyation, histone H3 Ly-9 methylation, and recruitment of HP1 proteins.
Interaction
TIF1β was shown to form homo-oligomers in vitro, but does not form hetero-oligomers with TIF1α or TIF1γ.
