    One week 4T1 cells (mouse breast cancer cell line) induced BALB/c frozen spleen section was fixed with 4% paraformaldehyde (PFA) for 10 minutes at room temperature and blocked with 5% FBS for 1 hour at room temperature. Then the section was stained with 5 µg/ml of CD3ε (clone 500A2) Alexa Fluor® 594 (blue), CD74 (clone In1/CD74) Alexa Fluor® 488 (green) and B220 (clone RA3-6B2) Alexa Fluor® 647 (red) at 4°C overnight. The image was scanned with a 10X object and stitched with MetaMorph® software.

Description

CD74, also known as CLIP, is a 33.5kD homotrimer and functions as a MHC class II chaperone for exogenous peptides and other MHC class II molecules. CD74 binds MIF and regulates innate and adaptive immunity through CD44 signaling. It is found primarily on B-cells, monocytes, macrophages and dendritic cells, but can also be found on activated T-cells, endothelial and epithelial cells.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤0.5 µg per million cells in 100 µl volume. For immunohistochemistry, a concentration range of 5.0 - 10 µg/ml is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 488 has a maximum emission of 519 nm when it is excited at 488 nm.
