Mouse serum (lane 1)and human serum (lane 2) (10µg protein for each) were resolved by 4-20% Tris-glycine gel electrophoresis, transferred to nitrocellulose, and probed with 1:1000 purified anti-human IgG Fc Antibody or 1:1000, 1:2500 and 1:10000 Direct-Blot™ HRP anti-human IgG1 Fc Antibody (upper). Proteins were visualized by chemiluminescence detection using a goat anti-rat-IgG secondary antibody conjugated to HRP. Ponceau S staining (lower) was used as loading control. Lane M is the Molecular weight ladder.

Description

IgG Fc is a homodimer that is composed of the constant region of the two heavy chains that form the IgG molecule. The Fc fragment mediates opsonization, antibody dependent cellular cytotoxicity (ADCC), and complement activation through the binding to Fc receptors such as CD16, CD32, CD64, and the complement factor C1.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested dilution is 1:1000-1:10,000. The optimal dilution should be determined by titration for each individual assay of interest.

25 µl and 100 µl of Direct-Blot™ HRP antibody can be used for approximately 5 and 20 Western blots, respectively, at the recommended concentration/dilution.
