Total lysates (15 µg protein) from HeLa (lane 1), Daudi (lane 2), and NIH3T3 (lane 3) cells were resolved by electrophoresis (4-12% Bis-Tris gel), transferred to nitrocellulose, and probed with 1:500 diluted (1 µg/mL) purified anti-c-REL Antibody, clone 2F5A22 (upper). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-mouse-IgG secondary antibody conjugated to HRP for the anti-c-REL Antibody. 1:5000 diluted Direct-Blot HRP anti-β-Actin Antibody (2F1-1) was used as a loading control (lower). Lane M: Molecular weight ladder.

    Chromatin Immunoprecipitation (ChIP) was performed using commercial Protein-G coated 96 well high-throughput ChIP assay kit by loading 3µg of cross-linked chromatin samples from Jurkat cells treated with TPA and Ionomycin with either A) 1:50 dilution of Go-ChIP-Grade™ Purified anti-c-REL (Clone 2F5A22), B) equal amount of Purified Mouse IgG2b, κ Isotype Control Antibody, or C) competitor’s ChIP-grade Purified anti-c-REL Antibody and D) equal amount of matched Isotype Control Antibody as recommended by the manufacturer. The enriched DNA was purified and quantified by real-time qPCR using primers targeting human Bcl2L1 gene region and human αSatellite repeat (as a negative control). The amount of immunoprecipitated DNA in each sample is represented as signal relative to the 5% of total amount of input chromatin.

Description

c-Rel is a member of the Rel/NF-κB transcription factor family that forms homo or heterodimers with family members such as RelA/p65, RelB, NF-κB1/p105, and NF-κB2/p100. The NF-κB complex is retained in cytoplasm by the inhibitor IκB protein in unstimulated cells. Upon activation, IκB is degraded via the ubiquitin-proteasome pathway, which results in nuclear translocation of the active NF-κB complex. While other NF-κB transcription factors are expressed in most cell types, relatively higher levels of c-REL expression are found in hematopoietic cells. c-REL is thought to play key roles in regulating proliferation, differentiation, and survival of B and T lymphocytes.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.5 - 2.0 µg per ml. For ChIP applications, the suggested dilution is 1:50-1:100 by volume. It is recommended that the reagent be titrated for optimal performance for each application.
