The cells were then stained with 5 µg/mL of the purified anti-human TNFRSF10B antibody (left) or purified mouse IgG2a, κ isotype control (panel B) overnight at 4°C followed by goat anti-mouse IgG (clone Poly4053) Alexa Fluor® 647 for an hour at room temperature. Nuclei were counterstained with DAPI. The image was captured with a 40X objective.

Description

DR5, also known as TRAIL-R2, TRICK2, KILLER, and CD262, is 55 kD member 10B of the TNF receptor superfamily (TNFRSF10B). It binds the cytotoxic ligand, TRAIL, and induces apoptosis. The DR5 receptor is broadly expressed on a variety of normal tissues and many tumors. DR5 expression has been reported to be upregulated in human cells by interferon-α, 2-methoxyestradiol, and paclitaxel, and downregulated by adenoviral E3 proteins.

Recommended Usage

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. For immunocytochemistry, a concentration of 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Clone A21121I does not block the staining of reference clone DJR2-4 (7-8)

Recombinant protein completely blocks the staining of test clone A21121I.
