p21 (also known as CIP1 or CDKN1A) is a G1 cell cycle regulator that inhibits cyclin-cyclin dependent kinase 2 and/or cyclin-cyclin dependent kinase 4 complexes. Its expression is highly controlled by the tumor suppressor protein p53, thus, p21 is involved in controlling the G1 cell cycle arrest in response to different stress stimuli. p21 plays a regulatory role in S phase DNA replication by interacting with PCNA (proliferating cell nuclear antigen). Activation of cyclin-dependent kinase 2 has been observed after p21 is cleaved by CASP3-like caspases. Loss of function of p21 has been associated with bladder cancer, Multiple Endocrine Neoplasia Type I and lymph node metastasis. In addition, p21 can bind the HIV integrase mediating resistance to infection in hematopoietic cells.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody (clone W22006E) in immunocytochemistry (ICC), we recommend fixation/permeabilization with either PFA/Triton X-100 or PFA/Methanol. This antibody is not suitable for use with methanol fixation/permeabilization.
