Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane and then probed with Purified anti-p21 (clone W22006E). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426318) was used as a detection agent. Lane M: molecular weight.

    IHC staining with Purified anti-p21 on formalin-fixed paraffin-embedded human colon tissue was performed following antigen retrieval using Tris-EDTA (10 mM Tris, 1 mM EDTA, 0.05% Tween-20, pH 9.0). The tissue sections were incubated without (panel A) or with Purified anti- p21 (clone W22006E) (panels B (in 10X) and panel C (in 40X)) overnight at 4°C followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801). Scale Bar: 50 µm

    Serum-starved A431 cells (negative control, top panels) and A549 cells (positive control, bottom panels) were fixed and permeabilized with 4% PFA and ice-cold methanol. They were then intracellularly stained with Purified anti-p21 (clone W22006E) (right panels) or Purified rat IgG2b, κ isotype control (Cat. No. 400602) (left panels). This was followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) (red). Nuclei were counterstained with DAPI (Cat. No. 425401) and intracellularly stained with Purified anti-p21 (clone W22006E) or Purified rat IgG2b, κ isotype control (open histogram, dashed line) (representative histogram for either cell line) (Cat. Description

p21 (also known as CIP1 or CDKN1A) is a G1 cell cycle regulator that inhibits cyclin-cyclin dependent kinase 2 and/or cyclin-cyclin dependent kinase 4 complexes. Its expression is highly controlled by the tumor suppressor protein p53, thus, p21 is involved in controlling the G1 cell cycle arrest in response to different stress stimuli. p21 plays a regulatory role in S phase DNA replication by interacting with PCNA (proliferating cell nuclear antigen). Activation of cyclin-dependent kinase 2 has been observed after p21 is cleaved by CASP3-like caspases. Loss of function of p21 has been associated with bladder cancer, Multiple Endocrine Neoplasia Type I and lymph node metastasis. In addition, p21 can bind the HIV integrase mediating resistance to infection in hematopoietic cells.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

For use of this antibody (clone W22006E) in immunocytochemistry (ICC), we recommend fixation/permeabilization with either PFA/Triton X-100 or PFA/Methanol. This antibody is not suitable for use with methanol fixation/permeabilization.
