Description

CD28 homolog (CD28H), also referred to as TMIGD2 (Transmembrane and Immunoglobulin Domain Containing 2) and IGPR-1 (Immunoglobulin-containing and Proline-rich Receptor-1), is a member of the immunoglobulin superfamily. It is predominantly expressed in epithelial and endothelial cells. The CD28H structure consists of an extracellular domain resembling an immunoglobulin V-like (IgV-like) domain, which contains two potential glycosylation sites. It also features a transmembrane segment and a cytoplasmic tail with a tyrosine residue. CD28H was previously recognized both as a co-stimulatory receptor and as an adhesion molecule. HHLA2 (B7-H7) is the known ligand for CD28H. Connection of the CD28H and HHLA2 primarily co-stimulates human T cell growth through an Akt-dependent signaling cascade which mediate tumor resistance against NK cells and promotes cytokine production. CD28H is associated with immune dysregulation and angiogenesis. In autoimmune diseases, abnormal CD28H signaling can cause inappropriate T cell activation, resulting in tissue damage. Additionally, CD28H expression can promote the abnormal self-renewal and differentiation of acute myeloid leukemia cells via downstream signaling pathways like ERK1/2-p90RSK-CREB signaling. Targeting CD28 signaling with anti-CD28 mAb attenuates self-renewal of leukemia stem cells, providing promising therapeutic value.

Biotinylated Recombinant Human CD28H, amino acid (Leu23-Gly150) (Accession # Q96BF3.2), with a linker, a C-terminal Human IgG (Pro100-Lys330), and an Avi-tag, was site-specifically biotinylated by enzyme BirA, was expressed in 293E.
Molecular Mass
The 380 amino acid recombinant protein has a predicted molecular mass of approximately 42.46 kD. The DTT-reduced and non-reduced proteins migrate at approximately 50 -70 kD and 75 – 120 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Leu.
Purity
> 95%, as determined by Coomassie-stained SDS-PAGE.

Concentration
25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling
Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin the vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity
When recombinant human B7-H7 is immobilized at 1 µg/mL, biotinylated recombinant human CD28H binds in a dose-dependent manner with ED50 range of 3 – 18 ng/mL. HRP-Avidin (Cat. No. 405103) was used to detect the binding.
