Description

The Myeloid Activation Analysis Cocktail facilitates rapid discrimination between viral and bacterial infections in a single simple test. This cocktail specifically helps to identify increased expression of CD169 on monocytes during viral infection and surface expression of CD64 on neutrophils in response to bacterial pathogens. It is optimized for use with a simplified No Wash staining protocol and analysis without the need for compensation.

The Myeloid Activation Analysis Cocktail contains:
PE anti-human CD169 (Sialoadhesin, Siglec-1) (clone 7-239)
APC anti-human CD64 (clone S18012C)
Spark Violet™ 423 anti-human HLA-DR (clone L243)

This cocktail contains PE, APC and Spark Violet™ 423 conjugated antibodies in an optimal concentration for flow cytometric analysis.
Preparation
All antibodies were purified by affinity chromatography and conjugated under optimal conditions.
Concentration
Lot-specific (to obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.)
Storage & Handling
The antibody solution should be stored undiluted between 2°C and 8°C, and protected from prolonged exposure to light. Do not freeze.
Application

FC - Quality tested

Recommended Usage

Each lot of the antibody cocktail is quality control tested. For flow cytometric staining, the suggested use of this reagent is 5 µL per 50 µL volume of whole blood. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Protocol - Whole Blood Lyse/No Wash Procedure:

  1. Prepare 1X RBC Lysis/Fixation solution by diluting 1 part of BioLegend RBC Lysis/Fixation solution (Cat. No. 422401) with 9 parts room temperature deionized water.
  2. Add 5 µL of the Myeloid Activation Analysis Cocktail to tube/well.
  3. Add 45 µL of Cell Staining Buffer (Cat. No. 420201) or equivalent.
  4. Mix in 50 µL of whole blood.
  5. Incubate at room temperature, in the dark, for 20 minutes.
  6. Add 0.45 mL of 1X RBC Lysis/Fixation solution, gently mixing each tube immediately.
  7. Incubate at room temperature, in the dark, for 15 minutes.
  8.  Analyze samples by flow cytometry.
