Total cell lysates (15µg protein) from HUVEC and HUVEC treated with recombinant Human TNF-α (10 ng/ml, 16 h) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to nitrocellulose, and probed with 1:25000 (0.02µg/ml), 1:50000 (0.01µg/ml) and 1:250000 (0.002µg/ml) purified anti-CD106 (VCAM-1) antibody (clone A16047A) or competitor’s antibody used at manufactures recommended concentration (upper). Proteins were visualized by chemiluminescence detection using a goat anti-mouse-IgG secondary antibody conjugated to HRP for clone A16047A, and a donkey anti-rabbit IgG Antibody conjugated to HRP for competitor’s antibody. Direct-Blot™ HRP anti-β-actin Antibody was used as a loading control (lower). Lane M is the Molecular weight ladder, M* indicates longer exposure.

Description

CD106 is a glycosylphosphatidylinositol (GPI)-linked transmembrane protein, also known as VCAM-1 and INCAM-110. It is constitutively expressed on bone marrow stromal cells, myeloid progenitors, splenic dendritic cells, activated endothelial cells, as well as some lymphocytes. CD106 expression can be upregulated on endothelial cells by inflammatory cytokines. CD106 is involved in adhesion and acts as a counter-receptor for VLA-4 (α4/β1 integrin) and LPAM-1 (α4/β7 integrin).

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.002 - 0.1 µg/mL (1:5000-1:250000 diluted). It is recommended that the reagent be titrated for optimal performance for each application.
