CDC73, also referred to as parafibromin or HRPT2, is a tumor-suppressor protein which plays a role in the suppression of epithelial-mesenchymal transition (EMT), inhibition of cell cycle progression and proliferation, and induction of apoptosis. In the nucleus, along with PAF1, LEO1, and CTR9, CDC73 forms the polymerase-associated factor (PAF) protein complex, which interacts with RNA polymerase II and the histone methyltransferase complex to control the expression of genes involved in cell cycle and metabolism, RNA polymerase II elongation, mRNA synthesis and processing, and chromatin and histone modifications. It also mediates IFN-γ signaling through interactions with JAK1 and JAK2. Loss of CDC73 function is associated with gastric carcinoma and mutations in CDC73 are linked to hyperparathyroidism-jaw tumor syndrome and parathyroid tumors. CDC73 plays a role in the coordinated activation and inactivation of Wnt, Hedgehog, and Notch signaling pathways through its role as a scaffold protein, where it sequesters the transcriptional factors β-catenin and GLI1. These interactions are dependent upon tyrosine phosphorylation of CDC73 mediated by PTK6 kinase or tyrosine dephosphorylation mediated by SHP2 phosphatase.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.0 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

This clone was tested for ICC using HeLa cells fixed with 4% paraformaldehyde and permeabilized with either methanol or Triton X-100. Both methods were compatible with CDC73 staining.
