    IHC staining of Claudin-6 was performed on formalin-fixed paraffin embedded human ovarian cancer tissue following antigen retrieval using Citrate buffer (Cat. No. 420902). The tissue sections were incubated with Purified anti-Claudin-6 (clone W22275B) (panel B) followed by Alexa Fluor® 647 Goat anti-rat (Cat. No. 405416) or secondary antibody only (panel A). Nuclei were counterstained with DAPI (Cat. No. 422801) (panel A and overlay in panel C). Scale Bar: 50 µm

    HeLa cells (negative cell line, panel A) and OVCAR-3 cells (positive cell line, panels B and C) were fixed with 4% PFA Fixation Buffer (Cat. No. 420801), permeabilized with 0.5% Triton X-100 and then blocked with 5% FBS. Cells were stained with Purified anti-Claudin-6 (clone W22275B) followed by Alexa Fluor® 647 Goat anti-rat (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801)(overlay in panels A and C). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Description

Claudin-6 is a member of the claudin family of proteins, which consists of at least 24 members. Claudin proteins are integral membrane proteins characterized by 4 transmembrane regions and 2 extracellular loops that form tight junctions. Abnormal expression can disrupt tight junction integrity, playing an important role in the occurrence and development of cancers. Claudin-6 in particular, has been considered a therapeutic target because it is commonly expressed in tumor tissue but not in healthy adult tissue.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 10 - 1 ug/mL is suggested. For immunocytochemistry, a concentration range of 10 - 1 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application. 

Application Notes

For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using PFA-Triton X-100 and Methanol. We do not recommend PFA-Methanol fixation/permeabilization.

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer, 10X (Cat. No. 420902), or Tris-EDTA buffer.
