Total cell lysates (15 µg total protein) from Daudi (reduced expression control), HeLa, SH-SY5Y, and NIH/3T3 cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a nitrocellulose membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-DDX3X antibody, clone 15D1B11, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 664804) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

    Immunoprecipitation of DDX3X from Hela cell extracts. Lane 1 is 5% input. Immunoprecipitation was performed using protein G resins only (lane 2), mouse IgG isotype control (lane 3), and anti-DDX3X antibody (clone 15D1B11, lane 4). Western blot was performed using anti-DDX3X antibody (clone 15D1B11). The astrostar indicates immunoglobulin heavy chain.

    Hela cells were fixed with 2% paraformaldehyde (PFA) for 10 minutes, permeabilized with 0.5% Triton X-100 for 5 minutes, and blocked with 5% FBS for 30 minutes. Then the cells were intracellularly stained with 2.5 µg/mL anti-DDX3X antibody (15D1B11) and followed by DyLight™ 594 goat anti-mouse IgG (red) for one hour at room temperature. Actin filaments were labeled with Alexa Fluor® 488 Phalloidin (green). Nuclei were counterstained with DAPI (blue). The image was captured with a 40X objective.

Description

DDX3X belongs to the DEAD (Asp-Glu-Ala-Asp)-box protein family of RNA helicases, involved in the alteration of RNA secondary structure required for transcription, mRNA processing, and translation initiation. DDX3X is an essential host factor during virus infection. DDX3X serves as the cellular target of Hepatitis C virus (HCV) core protein, and also facilitates nuclear export of immature HIV RNA. The RNA helicase activity of DDX3X is required for the replication of both HCV and HIV. DDX3X is implied to act as a viral RNA sensor, mediating antiviral innate immune signaling. DDX3 participates in IκB kinase ε (IKKε) and TANK-binding kinase 1 (TBK1) dependent IRF3 activation, leading to the production of Type I interferon. DDX3X was also proposed to have antiapoptotic functions and participate in tumorigenesis.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.25 - 1.0 µg per ml. For immunocytochemistry, the suggested use of this reagent is 1.0 - 5.0 µg per ml. For immunoprecipitation, the suggested use of this reagent is 5.0 - 20 μg per ml. It is recommended that the reagent be titrated for optimal performance for each application.
