Whole cell extracts (15 µg protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 0.1 µg/mL (1:5000 dilution) of purified anti-Fatty Acid Synthase (clone 2F9-B2-B6) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-actin antibody (Cat.     ICC staining of purified anti-Fatty Acid Synthase (clone 2F9-B2-B6) on HeLa cells. The cells were fixed and permeabilized with ice-cold methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 1.0 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG antibody (Cat. No. 405326) for 1 hour at room temperature. Nuclei were counterstained with DAPI, and the image was captured with a 60X objective.

    IHC staining using purified anti-Fatty Acid Synthase (clone 2F9-B2-B6) on formalin-fixed paraffin-embedded human breast tissue. The tissue was incubated with 10 μg/mL of the primary antibody overnight at 4°C, followed by incubation with 2.5 μg/mL of Alexa Fluor® 647 goat anti-mouse IgG antibody (red) (Cat. No. 405322) for one hour at room temperature. Nuclei were counterstained with DAPI (blue), and the slide was mounted with ProLong™ Gold Antifade Mountant. The image was captured with a 40x objective. Description

Fatty acid synthase is a lipogenic enzyme required for the terminal step of de novo fatty acid synthesis. Using NAPDH as a cofactor, it catalyzes the synthesis of palmitate from acetyl- and malonyl-CoA precursors. Fatty acid synthase is upregulated and serves as a prognostic marker in multiple cancers. In contrast to normal tissue, where the enzyme functions in anabolic lipid storage pathways, fatty acid synthase supports tumor growth by providing intermediates for membrane biosynthesis and provides alternative fuel sources for ATP production.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested dilution of this reagent is 1:2000. For immunocytochemistry, a dilution of 1:200 is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a dilution of 1:20 is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

During PD testing, additional bands below fatty acid synthase were observed. These were also produced by a control antibody raised with a different fatty acid synthase immunogen. We therefore believe these represent fatty acid synthase degradation products.

For ICC staining, we recommend using 4% PFA plus methanol or methanol only as fixation-permeabilzation methods. We do not recommend 4% PFA + Triton X-100 due to high non-specific nuclear staining when this method is used.
