ICC staining of purified anti-GSK3B (clone 9B1-5C3-5G8) on A431 cells. The cells were fixed and permeabilized with ice-cold methanol, and blocked with 5% FBS for 1 hour at room temperature. The cells were then stained with 2.5 µg/mL of the primary antibody, followed by incubation with 2.5 µg/mL of Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) for 1 hour at room temperature. Nuclei were then counterstained with DAPI (panel A) (Cat. No. 422801), or not counterstained (panel B), and the image was captured with a 60X objective. -
IHC staining of purified anti-GSK3B (clone 9B1-5C3-5G8) on formalin-fixed paraffin-embedded human testis tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with 5.0 μg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin HRP Kit (Multi-Species, DAB) (Cat. No. 929501) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 μm

GSK3B is a serine-threonine kinase and a member of the glycogen synthase kinase subfamily. It is a negative regulator of the hormonal control of glucose, Wnt signaling and also transcriptional factors and microtubules. It has been associated with regulation of several cellular mechanisms such as glycogen synthesis, protein synthesis, cell proliferation, microtubules stabilization, cell migration, autophagy and apoptosis. GSK3B is activated by phosphorylation at tyrosine 216 and inactivated by phosphorylation at serine 9. Defects on GSK3B has been associated with Alzheimer’s and Parkinson’s diseases.

IHC-P - Verified - Recommended Usage

Each lot of this antibody is quality control tested by immunocytochemistry. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 4.0 - 6.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

For ICC, we recommend 4% PFA fixation followed by permeabilization with ice-cold methanol, or fixation/permeabilization with ice cold methanol alone. We do not recommend 4% PFA fixation followed by permeabilization with 0.5% Triton-X.
For IHC-P, we recommend antigen retrieval using citrate buffer, HIER, and overnight antibody incubation at 4°C. - RRID
