IHC staining of anti-HCN1 antibody (clone N70/28) on formalin-fixed paraffin-embedded mouse eye tissue. Following antigen retrieval using Sodium Citrate H.I.E.R. (Cat. No. 928502), the tissue was incubated with 5.0 µg/mL of the primary antibody overnight at 4°C. BioLegend’s Ultra Streptavidin HRP Kit (Multi-Species, DAB, Cat. No. 929501) was used for detection followed by hematoxylin counterstaining, according to the protocol provided. The image was captured with a 40X objective. Scale bar: 50 μm.

Description

Hyperpolarization-activated cyclic nucleotide-gated (HCN) 1 channel is a member in the superfamily of six-transmembrane segment channels, and related to cyclic nucleotide-gated channels and voltage-dependent K+ channels. Four isoforms, HCN1-4, have been identified with up to 60% of sequence homology. The transmembrane α-helices and C-terminal cyclic-nucleotide binding domain of HCN channels are important for K+ ions selectivity filter and cAMP modulation. HCN1 is widely expressed in the nervous system where it plays a role in regulating membrane potential, dendritic integration, neuronal pacemaking, and action potential threshold.

IHC-P - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

Reactivity of clone N70/28 with human HCN1 is observed but weaker than reactivity with mouse or rat HCN1.
