Adenylate-uridylate-rich elements, or AREs, are short regions of RNA found in the 3’ UTR of messenger RNA. AREs are a major determinant of mRNA stability and translational efficiency, and play a crucial role in the post-transcriptional regulation of genes involved in several cellular processes.
In humans, the Hu family of proteins is comprised of 4 members, each of which stabilize ARE-containing mRNA through directly binding to the ARE motif. In contrast to the Hu family members HuB, HuC, and HuD, which show neural specific distribution, HuR (also known as ELAVL1) is expressed in all tissues. While HuR is largely a resident nuclear protein under normal conditions, in response to cellular stress it can chaperone mRNA from the nucleus to the cytosol and continue to stabilize mRNA once in the cytosol.
HuR is overexpressed and cytosolically delocalized in a wide range of cancers. In in vitro studies, HuR has been shown to stabilize several ARE-harboring mRNA encoding proteins involved in tumorigenesis and transformation, including MMP9, Bax, Fos, p21, and multiple cyclins, and is promising therapeutic target for cancer therapy.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 5.0 μg/mL is recommended. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

This clone was tested for ICC using 4% PFA-fixed HeLa cells permeabilized with either methanol or Triton X-100. While staining was observed with both methods, methanol permeabilization resulted in stronger HuR staining.
