Whole cell extracts (15 µg protein) from the indicated cells were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 0.25 µg/mL (1:2000 dilution) purified anti-KEAP1 antibody (clone W19335C) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular weight marker. Cell lines were loaded in order of increasing predicted KEAP1 expression (source: Human Protein Atlas).

Description

NRF2 is a transcription factor that plays a critical role in inducing expression of genes required for oxidative stress defense and stress balance. Active NRF2 binds to antioxidant response elements (AREs) located in the regulatory promoter of target genes and induces expression of these genes. NRF2 is degraded through ubiquitination by an E3 ligase complex that contains KEAP1 as an adaptor protein. Under normal conditions, KEAP1 directly binds and sequesters NRF2 in the cytosol and promotes its ubiquitinatation and proteasomal degradation. In response to elevated oxidative stress, reactive cysteine residues on KEAP1 are modified, resulting in a conformational change that causes the dissociation of NRF2 from KEAP1, and an accumulation of NRF2 in the nucleus. In a clinical setting, NRF2 is frequently activated in many types of cancers, driving metabolic adaptation and survival in ROS-rich tumor microenvironments. In KRAS-mutant cancers, loss of KEAP1 promotes dependence on glutaminolysis through upregulation of NRF2.
