CD91, also known as LRP1, is a transmembrane endocytic and cell signaling protein that regulates diverse cellular functions in the brain, liver, kidney, lung, and vasculature. The alpha and beta chains of LRP1 form a transmembrane receptor on the plasma membrane that plays a role in receptor-mediated endocytosis and lipoprotein metabolism. LRP1 gene encodes a 4545 amino acid precursor protein that is processed in the trans-Golgi complex into the 505 kD. It is involved in phagocytosis of apoptotic cells and cellular liquid homeostasis, as well as the plasma clearance of chylomicron remnants and activated LRPAP1 (alpha 2-macroglobulin). CD91 is also essential to the local metabolism of complexes between plasminogen activators and their endogenous inhibitors. CD91 acts as an alpha-2-macroglobulin receptor and TAU/MAPT receptor and controls the endocytosis of TAU/MAPT as well as its subsequent spread and is required for early embryonic development.
