IHC staining of purified anti-NAC-1 antibody (clone SWN-3) on formalin-fixed, paraffin-embedded human brain cortex tissue. Following antigen retrieval using sodium citrate, the tissue was incubated with 10 µg/mL of the primary antibody for 24 hours at 4°C. BioLegend's Ultra Streptavidin (USA) HRP Detection Kit was used for detection, follow by counterstaining with hematoxylin.

    Western blot of purified anti-NAC-1 antibody (clone SWN-3). Lane 1: molecular weight marker, Lane 2: 20 µg of SH-SY5Y cell lysate. The blot was incubated with the primary antibody at 5 µg/ml overnight at 4°C, followed by incubation with HRP-labeled goat anti-mouse secondary antibody. Enhanced chemiluminescence was used as the detection system.

Description

NAC-1 is a member of the POZ/BTB protein family involved in several cellular processes including proliferation, apoptosis and transcription regulation. NAC-1 is required for recruiting the proteasome from the nucleus to the cytoplasm and dendritic spines.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 5.0 - 10 µg/ml is suggested. For Western blotting, the suggested use of this reagent is 1.0 - 10 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

NAC-1 is a 527 amino acid protein with an apparent molecular mass of 57.3 kD.
Distribution

Tissue distribution: brain, ovary, testis, and thyroid.
Cellular distribution: nucleus and cytosol.

Function
NAC-1 is a transcriptional repressor that plays a role in stem cell self-renewal and pluripotency maintenance. NAC-1 also contributes to the progression of multiple types of cancer by suppressing the transcription of the tumor suppressor Gadd45GIP1. It also functions as a transcriptional corepressor in neuronal cells through recruitment of HDAC3 and HDAC4.
