Total cell lysates (15 µg protein) from HeLa (lane 1), PC3 (lane 2), Jurkat (Lane 3) and NIH3T3 cells (lane 4) were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with 1:500 diluted (1 µg/mL) Purified anti-Pax9 Antibody, clone 14D10A24 (upper panel). Proteins were visualized by chemiluminescence detection using a 1:3000 diluted goat anti-mouse-IgG secondary antibody conjugated with HRP for anti-Pax9 Antibody or 1:5000 diluted Direct-Blot HRP anti-β-Actin Antibody, clone 2F1-1(lower) as a loading control. Lane M: Molecular weight ladder.

Description

PAX9 is a transcription factor belonging to the PAX family, which is composed of nine members defined by the presence of a paired box domain responsible for DNA binding. PAX9 is expressed at the E10 stage and downregulated at the bell stage. It is controlled by Fgf/Bmp signaling and plays an essential role in tooth development. PAX9 deficient mice fail to form teeth and show developmental arrest at the bud stage. In humans, heterozygous mutation in PAX9 is associate with tooth agenesis. These findings support the role of PAX9 in tooth development.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This antibody binds non-specifically to a protein over 250kDa therefore it is not recommended to use in immunofluorescence staining.
