Whole cell extracts (15 µg protein) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of purified anti-PKCι antibody (clone W18205A) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH Antibody (Cat. No. 607904) was used as a loading control at a 1:25000 dilution (lower). Lane M: Molecular weight marker. Cell lines were loaded in order of ascending PRKCI expression; predicted expression data obtained from Human Protein Atlas.

    Whole cell extracts (250 µg total protein) prepared from HeLa cells were immunoprecipitated overnight with 2.5 µg of Purified Rat IgG2a, κ Isotype Ctrl Antibody (Cat. No. 400602) or purified anti-PKCι antibody (clone W18205A). The resulting IP fractions and whole cell extract input (10%) were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with 1.0 µg/mL (1:500 dilution) of clone W18205A. The secondary step was performed using light-chain-specific secondary antibodies. Lane M: Molecular weight marker.

Description

Members of the protein kinase c family (PKC) are serine/threonine protein kinases with a broad set of substrates that can regulate diverse cellular processes. PKCι, also referred to as PKCι/λ, is a member of the atypical subfamily of PKC, which is characterized by complete independence of calcium and diacylglycerol as second messengers. It has been shown to play a role in cancer survival through multiple mechanisms: a. phosphorylation of the DNA-binding domain of FoxO1 resulting in transcriptional reprogramming of c-myc, which leads to increased proliferation of angiosarcoma cells, b. regulating membrane trafficking of type 1-matrix metalloprotease from late endosomal compartments to the plasma membrane, and c. mediating pro-survival by phosphorylating and inhibiting the pro-apoptotic factor BAD in glioblastomas. PKCι is also required for resistance to anti-apoptotic drugs in BCR-Abl-dependent leukemia and may represent a viable target for combinatorial therapies.

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. For immunoprecipitation, the suggested use of this reagent is 2.5 µg/test. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone was tested for ICC using HeLa cells fixed with 4% PFA and permeabilized with methanol or Triton X-100; it failed due to weak staining.
