Tetraspanins, also referred to as the transmembrane 4 superfamily (TM4SF) proteins, have four transmembrane alpha-helices and two extracellular domains. It acts as molecular facilitators forming a web between themselves and a multitude of molecules. It is also involved in adhesion, motility, matrix modulation and cell fusion. Tetraspanins/Tetraspanin-complexes are constitutive components of exosomes. During exosome assembly, Tetraspanins become internalized via a tyrosine-based internalization motif. Internalized membrane microdomains are recruited into early endosomes from where they can become incorporated into multivesicular body (MVB), which migrate towards and fuse with the cell membrane releasing invaginated endosomes as exosomes.

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 1.0 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
