Whole cell extracts (15 µg total protein) from HeLa (reduced expression control), Daudi, Ramos, and Ba/F3 cells were resolved by 4-12% Tris-Glycine gel electrophoresis, transferred to a nitrocelullose membrane, and probed with 2.0 µg/mL (1:250 dilution) of Purified anti-RelB Antibody, clone W16195A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG Antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-β-Actin Antibody (Cat. No. 664804) was used as a loading control at a 1:10000 dilution (lower). Lane M: Molecular Weight marker. Predicted expression data was obtained from Human Protein Atlas.

Description

The Nuclear Factor kappa-light chain-enhancer of activated B cell, or NF- κB, is a transcriptional complex that plays a critical role in inducing expression of genes involved in inflammation, stress responses, cell growth, and apoptosis. The functional NF- κB complex can be comprised of homo- or heterodimeric units of Rel-like/homology domain containing proteins, including RelA, RelB, cRel, p50 and p52. RelB, which preferentially forms heterodimers with p50 and p52, is associated with non-canonical NF- κB signaling activation. This branch of NF- κB signaling is characterized by the NF-κB-inducing kinase (NIK)-linked processing of p100 instead of degradation of IκBα. Non-canonical NF-κB signaling is crucial for B-cell survival and maturation, and overexpression of RelB is associated with lymphoid malignancies and colorectal tumorigenesis.

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.125 - 2.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone may produce a ~65 kD non-speccific band by WB when used at higher concentrations.

This clone does not recognize mouse RelB.
