Whole cell extracts (15 µg) from the indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane and probed with a 1.0 µg/mL (1:500 dilution) of purified anti-SEC13 antibody, clone W18034A, overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP-conjugated goat anti-rat IgG antibody (Cat. No. 405405) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH antibody (Cat. No. 607904) was used as a loading control at a 1:50000 dilution (lower). Lane M: Molecular Weight marker.

Description

SEC13 functions as a component of the nuclear pore complex (NPC) involved in nucleo-cytoplasmic trafficking. At the ER, SEC13 is involved in the biogenesis of COPII-coated vesicles, which are required for protein export from the ER to either the Golgi complex or the ER-Golgi intermediate compartment (ERGIC). It also functions as a component of the GATOR2 complex, which acts as an amino acid-sensing branch of the TOR1 signaling pathway.

WB - Quality tested

Recommended Usage

Each lot of this antibody is quality control tested by Western blotting. For Western blotting, the suggested use of this reagent is 0.05 - 1.0 µg per ml. It is recommended that the reagent be titrated for optimal performance for each application.

Application Notes

This clone failed IP testing when the target was pulled down and probed via WB.

In addition, a nonspecific band was detected in the initial Western Blot screen; however, that band was titrated out at ~0.1 µg/mL. Nevertheless, the presence of the band indicates that the clone is not suitable for ICC testing.
