Whole cell extracts (15 µg total protein) from the indicated cell lines were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with 1 µg/mL (1:500 dilution) of purified anti-SKP1 (clone P84E11C10) overnight at 4°C. Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG (Cat. No. 405306) at a 1:3000 dilution. Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control at a 1:50000 dilution. Western-Ready™ ECL Substrate Plus Kit (Cat. No. 426317) was used as a detection agent. Lane M: Molecular weight marker -
HeLa cells were fixed and permeabilized with ice cold MeOH for 10 minutes, and blocked with 5% FBS for 60 minutes. Cells were then intracellularly stained with 5.0 µg/mL of either purified mouse IgG1, κ isotype control (Cat. No. 400102) (panel A) or purified anti-SKP1 (clone P84E11C10) (panel B) overnight at 4°C, followed by incubation with Alexa Fluor® 594 goat anti-mouse IgG (Cat. No. 405326) at 2.0 µg/mL. Nuclei were counterstained with DAPI and the image was captured with a 60X objective. Scale bar: 50 µm

S-Phase Kinase Associated Protein 1 (SKP1) is one of the core components of the Skp, Cullin, F-box containing (SCF) E3 ubiquitin ligase complex which poly-ubiquitinates protein targets for proteasomal degradation, and thereby regulates a variety of cellular processes including DNA damage response, apoptosis, cell cycle, and overall genome stability. Within the SCF, SKP1 serves as an adaptor protein linking the scaffolding protein Cul1 to F-box proteins. Beyond its role as part of SCF, SKP1 also functions in centrosome organization, chromosome segregation, meiotic progression, and oocyte survival. Aberrant SKP1 expression is associated with chromosomal misalignment and instability, disrupted cell cycle, and is a characteristic of several cancers.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 2.5 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For ICC, we recommend fixation with MeOH. Fixation in 4% PFA followed by permeabilization with MeOH, and fixation in 4% PFA followed by permeabilization with 0.5% Triton X-100 were both tested but produced weak staining.
This clone is not recommended for IHC-P. Testing was conducted using formalin-fixed paraffin-embedded human testis but only a faint signal was produced - RRID
